Well, here is the main point. I have 5.4k reserve stam to burn up, still have my apple from the cows and level 182 I think... I need to know what level would be the best time to use this up the most efficiently and what buffs to use.
Bonfire planning...

Hard to say what level to use it. To make it save you a bit of stam you should find a 2hit level where you fight in Armor set up and where you have to change gear (different from the best damage set up) to get enough armor to survive, and where you cant 1hit with SK.
Using the potion you will need less armor, so if you can change gear and reach enough damage to 1hit with SK, SA, MES, PS, you will save some stam.
Other than that it can probably save some imps, but not a huge benefit.
Probably more useful in other aspects of the game other than leveling.

At my current level, well at least my last hunts, I could 1 hit kill all the creatures in my realms without any buffs. But the problem there is isn't there a gear change coming up? I don't want to waste my time hunting down gear to hop into.

I'd say that you will go on one hitting for a long time, so to use that potion for hunting wont help much (if not giving some safe 1hitting, protected by armor). Game have changed too much since i hunted in your levels, but i know i havent used a 2hit armor set from 200ish (using Tarbidas, but no one in their right mind use it anymore  ) to around lvl600 with Polar.
In GvG could be a great potion though.
